Sr. Scientist/Principal Scientist - Bioanalysis

Cambridge, MA

We are seeking a highly motivated and skilled Sr. or Principal Scientist with expertise in LC/MS-based analysis of oligonucleotides to join our growing research team. The ideal candidate will have hands-on experience with mass spectrometry-based structural elucidation, including metabolite identification (Met ID) and preferably, in vitro hepatocyte cell culture systems. This individual will play a key role in supporting both discovery and development-stage programs through advanced analytical method development, sample analysis, and data interpretation.  Please note this is an onsite opportunity.

Responsibilities

  • Develop, optimize, and qualify LC/MS methods for qualitative and quantitative analysis of oligonucleotide-based therapeutics.
  • Design and execute in vitro hepatocyte incubation studies to evaluate metabolic stability and biotransformation pathways.
  • Utilize high-resolution mass spectrometry (HRMS) for sequence verification, metabolite profiling and protein biomarker analysis.
  • Perform structural characterization and metabolite identification (Met ID) of oligonucleotides and related compounds.
  • Collaborate closely with cross-functional teams in medicinal chemistry, DMPK, and biology to support project objectives.
  • Analyze, interpret, and present experimental data clearly and concisely to key stakeholders.
  • Ensure all work complies with regulatory guidelines, standard operating procedures (SOPs), and data integrity principles.
  • Prepare Bioanalysis and ADME reports for IND/CTA filing.

Qualifications

  • Ph.D. in Analytical Chemistry, Biochemistry, Pharmaceutical Sciences, or a related field with 5+ years of relevant industry experience; or M.S. with 8+ years of industry experience.
  • Deep expertise in LC-MS/MS and HRMS techniques for biomolecule characterization, especially oligonucleotides.
  • Experience in antisense oligonucleotide (ASO), siRNA, or mRNA therapeutic platforms.
  • Proficiency with mass spectrometry software (e.g., Xcalibur, MassLynx, Compound Discoverer, Analyst, etc.).
  • Demonstrated ability to troubleshoot complex analytical challenges and drive results.
  • Excellent organizational, communication, and documentation skills.

Preferred Qualifications

  • Proven experience in oligonucleotide metabolism, Met ID, and in vitro ADME studies using hepatocytes or similar systems.
  • Experience with LC/MS-based peptide and protein quantification
  • Knowledge of regulatory bioanalytical guidelines (FDA, EMA) is a plus.
