Scientist / Sr. Scientist, Library-Based Protein Engineering
Manifold Bio is a biotech company pursuing a pipeline of protein therapeutics using novel molecular measurement technologies and library-guided protein engineering. Our drug discovery engine is differentiated by massively parallel screening in vivo from the beginning of our discovery process. This unique platform is powered by a proprietary protein barcoding technology that allows multiplexed protein quantitation at unprecedented scale and sensitivity. We combine this and other high-throughput protein engineering approaches with computational design to create antibody-like drugs and other biologics. Position
We are seeking an exceptional scientist with deep library-based engineering expertise and a mix of wet lab and computational experience to join our team. In this role, you will go end-to-end on library-based projects to engineer therapeutic proteins. You will develop and execute novel high-throughput screening assays, design and construct DNA libraries using advanced cloning techniques, and analyze complex library screens, including massively parallel in vivo screens powered by our proprietary mCode protein barcoding technology. You will work with a highly interdisciplinary team of creative experimental, computational, and hybrid scientists to advance high-throughput protein engineering and synthetic biology initiatives. Requirements:
- PhD or equivalent experience in protein biochemistry, molecular biology, bioengineering, computational biology, or a related field
Wet lab expertise:
- Proficiency in standard molecular biology and biochemistry methods
- Hands-on experience with DNA library design, cloning, and selection systems
- Proven capability in library-based protein engineering, including experience with high-throughput or multiplexed assay formats
- Skilled in designing, executing, and analyzing complex experimental workflows from end to end
Computational biology and bioinformatic expertise:
- Proficiency in analysis of large data sets, ideally using Python
- Experience working with NGS data and DNA/protein sequence analysis
Core Competencies:
- Detail-oriented with exceptional organizational and communication skills.
- Curious and flexible, with a strong desire to work collaboratively within a fast-paced and multidisciplinary team environment.
Preferred Qualifications:
- 2+ years of start-up, biotech, or pharma experience in protein engineering or assay development.
- A love for tinkering with biological parts and solving challenges to develop novel assays.
- Enthusiasm for combining rational design with unbiased approaches to problem-solving.
Direct experience with:
- engineering antibodies including complex multispecific formats
- designing antibody or other therapeutic binder libraries
- display technologies and selections including phage, yeast, and/or ribosome display
- screening-based methods with an NGS readout, e.g. DMS, MPRA, CRISPR screening, cancer screens, single cell sequencing
- computational protein engineering methods, machine learning, or biophysical modeling
- developing or engineering brain shuttle technologies for improved blood-brain barrier transport
