Scientist, Antibody Engineering
Manifold Bio is a dynamic biotech company building a pipeline of targeted biologics using a novel in vivo-centric discovery approach. Our drug discovery engine is differentiated by massively parallel screening in vivo from the beginning of our discovery process. This unique platform is powered by a proprietary protein barcoding technology that allows multiplexed protein quantitation at unprecedented scale and sensitivity. We combine this and other high-throughput protein engineering approaches with computational design to create antibody-like drugs and other biologics. Our world-class team of protein engineers, biologists, and computational scientists are working together to aim the platform at therapeutic opportunities where precise targeting is the key to overcoming clinical challenges.
Position
Manifold Bio is seeking a creative and driven Scientist, Antibody Engineering to join our protein engineering team and advance the next generation of targeted therapeutics. In this role, you will drive comprehensive antibody screening and optimization programs from initial characterization through therapeutic development. You will execute sophisticated library-based antibody characterization methods, leverage our cutting-edge proprietary mCode technology for massively parallel in vivo screening, and lead affinity maturation efforts to optimize binding kinetics and other critical therapeutic properties of our antibody candidates.
You'll serve as a key contributor to our therapeutic pipeline, generating critical datasets that inform cross-functional program teams and support both internal therapeutic development and strategic partnership initiatives. You will work with a highly interdisciplinary team of creative experimental, computational, and hybrid scientists in a culture that embraces scientific boldness and takes a team-focused approach to solving the most challenging problems in targeted drug delivery.
This position offers the opportunity to work at the forefront of antibody engineering, combining established characterization methods with revolutionary in vivo screening technology to develop the next generation of precision therapeutics.
Responsibilities
- Execute library-based as well as more standard antibody characterization workflows
- Design and lead in vivo screens leveraging Manifold's proprietary mCode protein barcoding technology to identify antibodies with optimal tissue targeting capabilities
- Lead affinity maturation campaigns to optimize binding kinetics, specificity, stability, and other critical therapeutic properties through rational design and selection approaches
- Develop and optimize high-throughput screening assays and experimental workflows to support antibody discovery and engineering programs across multiple therapeutic targets
- Analyze complex screening datasets and collaborate with cross-functional teams to translate experimental findings into actionable insights for therapeutic program advancement.
Required Qualifications
- PhD or equivalent experience in protein biochemistry, molecular biology, biological engineering, or a related field
- 2+ years industry experience in therapeutic antibody engineering including affinity maturation and library-based biomolecule engineering (e.g. phage display, yeast display)
- Proficiency in standard molecular biology and biochemistry techniques, (cloning, protein characterization)
Preferred Qualifications
- Yeast display
- Developability characterization and engineering
- Computational skills, experience working with NGS data and DNA/protein sequence analysis
- Experience working with bispecific or multispecific antibodies, format engineering
- Experience working on brain shuttles
- Experience with a range of display technologies and selections including phage, yeast, and/or ribosome display
- Familiarity with SPR/BLI
- Experience with computational protein design or engineering methods Experience with automation and high-throughput screening
