Associate Scientist/Senior Associate Scientist, Protein Sciences

Boston

Manifold Bio is a dynamic biotech company building a pipeline of  targeted biologics using a novel in vivo-centric discovery approach. Our drug discovery engine is differentiated by massively parallel screening in vivo from the beginning of our discovery process. This unique platform is powered by a proprietary protein barcoding technology that allows multiplexed protein quantitation at unprecedented scale and sensitivity. We combine this and other high-throughput protein engineering approaches with computational design to create antibody-like drugs and other biologics. Our world-class team of protein engineers, biologists, and computational scientists are working together to aim the platform at therapeutic opportunities where precise targeting is the key to overcoming clinical challenges.

Position

We are seeking a highly motivated and enthusiastic individual to be an integral part of our protein science team. This person will be responsible for expressing, purifying, and characterizing novel multispecific antibody drug formats for in vitro and in vivo experiments, while working closely with CROs to supplement production capabilities. They will work closely with a collaborative team of scientists to design, execute, and analyze complex experiments in a fast-paced environment. There will also be many opportunities for creativity and innovation as we push the boundaries of our platform’s multiplexing technology in every step of the drug discovery process.

Responsibilities

  • Maintain mammalian cell cultures and perform transient transfections to produce protein therapeutic candidates
  • Purify antibodies and other proteins using AKTA FPLC systems
  • Optimize purification processes using affinity, ion exchange, hydrophobic interaction, and size exclusion chromatography
  • Develop processes for optimized protein expression and purification
  • Characterize proteins using techniques such as SDS-PAGE, HPLC-SEC, DLS, and UV-Vis spectrophotometry
  • Test binding interactions using biolayer interferometry
  • Work with and manage CROs conducting protein production
  • Plan and perform experiments, analyze data, and present results in group meetings
  • Document experiments in an electronic lab notebook

Required Qualifications

  • BSc or MSc and 5+ years of industry experience
  • Expertise in mammalian protein expression and purification
  • Strong proficiency with AKTA FPLC systems
  • Knowledge of antibody purification techniques
  • Hands-on experience with secondary purification techniques such as IEX, HIC and MMC
  • Proficient with basic protein characterization methods such as SDS-PAGE, HPLC, UV-Vis
  • Hard-working, scientifically curious, and enthusiastic to contribute to cutting-edge work
  • Strong verbal and written communication skills
  • Detail-oriented and able to multitask
  • Able to work well as part of a team

Preferred Qualifications

  • Experience managing CROs
  • Experience with expression optimization
  • Experience with purification optimization
  • Knowledge of DoE
  • Experience with high throughput protein production
  • Competency in protein characterization techniques such as SPR, BLI, DSF, DSC, cIEF, and CE-SDS

This Role Might Be Perfect For You If

You are curious and passionate about science and drug discovery and want to work with a fast-moving team that values scientific boldness and innovation. This role requires deep technical expertise combined with a willingness to work thoughtfully and quickly to solve scientific challenges in a collaborative manner.
